Somatic Embryogenesis and Plant Regeneration in Cotyledon Culture of Paeonia albiflora


Abstract
This study was carried out to investigate effects of plant growth regulators on somatic embryogenesis. Effect of low temperature pretreatment and culture temperature on plant regeneration from the somatic embryos was also investigated. Somatic embryogenesis occurred the most in the MS medium supplemented with 0.2 §·/L 2,4-D and 0.2 §·/L BA. Somatic embryos were directly formed on the surface of cotyledon explant. Germination of the somatic embryos was promoted by low temperature pretreatment at 1 ¡É for 45 days. When the embryos were in vitro cultured on MS medium supplemented with 0.5 §·/L BA and 1.0 §·/L GA©ý at 18 1¡É, rate of normal plantlet formation increased slightly.
